چکیده

Degradation products of nicotinamide adenine dinucleotide phosphate, formed under the hot mild alkaline conditions that are used in the determination of NADPH in the presence of NADP+, were found to inhibit the enzyme used for NADPH estimation. The agent primarily responsible for inhibition was isolated and identified as phosphoadenosine diphosphate ribose. Phosphoadenosine diphosphate ribose was found to be a very specific inhibitor for NADP+ dehydrogenases. It had no effect on NAD+-specific enzymes.

The ADP-ribosyl cyclases--the current evolutionary state of the ARCs.

The major ADP-ribosylating enzyme families are the focus of this special issue of Frontiers in Bioscience . However, there is room for another family of enzymes with the capacity to utilize nicotinamide adenine dinucleotide (NAD): the ADP-ribosyl cyclases (ARCs). These unique enzymes catalyse the cyclization of NAD to cyclic ADP ribose (cADPR), a widely distributed second messenger. However, th...

Studies on the Polymer of Adenosine Diphosphate Ribose

A particulate fraction from rat liver nuclei catalyzes polymerization of the adenosine diphosphate ribose moiety of nicotinamide adenine dinucleotide into an acid-insoluble polymer with the simultaneous release of nicotinamide. The polymer, prepared by reaction in vifro, can be phenolextracted and gives a single symmetrical peak upon equilibrium cesium sulfate density gradient centrifugation at...

Past and current topics on ADP-ribosylation reactions.

The milestone of Adenosine Diphosphate-ribosylation studies was the paper by Paul Mandel's group in 1960s, first describing a "sort" of polyadenylic acid synthesized upon addition of nicotinamide mononucleotide in rat liver nuclear extracts. Nicotinic Acid or Niacin is the precursor of Nicotinamide Adenin Dinucleotide. In 1960s this compound was known mainly as coenzyme of most redox processes ...

NAD⁺ in aging, metabolism, and neurodegeneration.

Nicotinamide adenine dinucleotide (NAD(+)) is a coenzyme found in all living cells. It serves both as a critical coenzyme for enzymes that fuel reduction-oxidation reactions, carrying electrons from one reaction to another, and as a cosubstrate for other enzymes such as the sirtuins and poly(adenosine diphosphate-ribose) polymerases. Cellular NAD(+) concentrations change during aging, and modul...

From toxins to mammalian enzymes: the diverse facets of mono-ADP-ribosylation

The ADP-ribosylation of proteins is a phylogenetically ancient mechanism that involves the transfer of ADP-ribose from nicotinamide adenine dinucleotide (NAD+) to specific amino acids of target proteins post-translationally. In the first part of this review, we briefly describe ADP-ribosylation as the mechanism of action of toxins, while giving particular emphasis to a non-conventional ADPribos...
